An immunised animal has already solved the hard problems of affinity, specificity and self-tolerance. Antibody discovery from an immune repertoire is therefore a sampling problem, and every stage of a campaign is a chance to miss the clone you were looking for.
This session walks through the design decisions behind a single B cell discovery platform, from a CRO perspective where the balance between quality, speed and cost is critical. It addresses the most valuable information to acquire before each irreversible step in the campaign.
Topics include immunisation design as the first decision, drawing on aggregated data from several hundred legacy campaigns; why the memory B cell and plasma cell compartments hold functionally different repertoires, and how regimen and harvest timing shift the balance; how to match screening technology to scale, target class and required readout, including cell-based assay formats for conformational and multi-pass membrane targets; why sorted events are not hits, and the cost of taking screening output straight into production; and how single B cell output feeds multispecific format scouting and developability selection through mammalian display.
